This didactive video explains the cryopreservation process that scientists at the CIP genebank developed and validated for the long-term conservation of potato germplasm. It shows step by step how potato can be conserved for decades, or centuries, in liquid nitrogen, without any need for further manipulation. Whenever needed, potato shoot tips that have been cryopreserved using this protocol can be thawed and, within 50-55 days, the recovered plants will be ready for transplanting to a greenhouse. The CIP cryobank currently conserves more than 4,300 potato accessions under high quality standards, making it the largest cryopreserved collection of any clonal crop in the world. External reviewers have called the CIP genebank a center of excellence for plant cryopreservation and it will soon operate as a cryohub for safety duplication of the clonal and intermediate seed crops preserved by Latin America’s largest genebanks. CIP has already begun providing capacity building and support for staff at those genebanks. The cryopreservation protocol shown here can serve as a model for other crops, and the aim of this video is to support other genebanks, research institutes, universities, and scientists interested in cryopreserving clonal crops. To the best of our knowledge, this is the first time the complete cryopreservation process has been shown in a video of this kind.
